艾拉伦是一家生物制药公司，致力于发现、开发与应用“RNAi核糖核酸干扰机转”治疗遗传性疾病。艾拉伦成立于2002年，总部位于美国麻塞诸塞州剑桥市，2016年曾入围福布斯“100家最具创新力的成长型公司”名单。^[3]

历史沿革

艾拉伦是由科学家Phillip Sharp、Paul Schimmel、David Bartel、Thomas Tuschl、Phillip Zamore，还有投资人Christoph Westphal与John Kennedy Clarke于2002年从Max Planck生物物理化学研究所拆分成立，John Maraganore为创始CEO。^{[4][5][6][7][8]}艾拉伦是以猎户座腰带上的一颗恒星-- Alnilam命名，后来为了公司的独特性，而改变拼写。^[9]

2003年，艾拉伦与德国制药公司Ribopharma AG合并，并从私募股权基金获得2460万美元的资金。^{[10] [11]}

2004年，2月27日艾拉伦申请了首次公开招股，^[12]共筹集了2600万美元，并开始以ALNY的名义在纳斯达克证券交易所进行交易。^[13]

2005年，与美敦力公司(Medtronic)合作共同开发复合性药物(药品/医材结合)，治疗神经退化性疾病。^[14]

2006年，与百健公司(Biogen Idec)合作，开发治疗进行性多灶性白质脑病(PML)的方法。^[15]

2007年，与Hoffmann-La Roche建立了一个非排他性联盟，其中艾拉伦以其技术平台的使用权换取3.31亿美元^[16]；同年，与Ionis制药公司合作成立了Regulus Therapeutics公司，专注于小分子核糖核酸(microRNA)疗法。^[17]

2009年，艾拉伦与Cubist Pharmaceuticals和Kyowa Hakko Kirin结为联盟，销售一款针对呼吸道融合细胞病毒的药物。^[18]

2010年，扩大之前与美敦力医疗产品公司的合作，将CHDI基金会加入亨丁顿舞蹈症的重点研究中。^[19]

2011年，与葛兰素史克公司(GlaxoSmithKline)合作开发核糖核酸干扰疗法(RNAi)，以增强疫苗生产。^[20][21]

2012年，与孟山都公司(Monsanto)建立了为期10年的联盟，通过开发用于作物保护的天然分子，为农业开发生物技术提供解决方案。^[22][23][24]同年，与赛诺菲健赞公司(Sanofi Genzyme)建立了合作关系，共同开发治疗淀粉样多发性神经病变，为一种发生在亚洲的遗传性疾病^[25]。

2013年2月，与麦迪逊医药公司(Medicines Company)进行合作，开发治疗高胆固醇遗传形式的药物^[26]。7月，第一期临床试验中数据显示，艾拉伦证明了可以将甲状腺素运载蛋白(transthyretin, TTR)明显减少，并证明透过静脉注射和皮下给药方式对人体的疗效^[27]。

2014年，赛诺菲健赞公司以7亿美元收购了艾拉伦12%的股份，并得到了艾拉伦几种药物的权利。另外，艾拉伦也宣布以2500万美元的现金和1.5亿美元的股票收购默克药厂的Sirna Therapeutics公司^[28][29]。

2015年，艾拉伦收入为4100万美元，市值为52亿美元^[30]。

2016年，艾拉伦总公司在马萨诸塞州诺顿市购买了土地，以建立一个生产设施。^[31][32]10月，产品revusiran的第三期临床试验由于试验中药物组的死亡人数增加而被停止，终止该化合物的开发。^[33][34]

2019年，来台湾登记成立分公司。^[35]

2020年2月，艾拉伦总公司任命前赛诺菲健赞基因公司首席执行官Olivier Brandicourt为董事会成员，^[36]同年正式在台湾设立办公室 。^[37]

产品

2016年，艾拉伦制药在遗传医学、心脏代谢疾病和肝脏感染性疾病方面，共有18种处于不同开发阶段的潜在治疗方法。^[38]

2018年，药物Onpattro（patisiran）成功获得美国FDA核准^[39]，并于2020年也获得台湾食药署核准^[40]，为美国FDA跟台湾食药署首支批准的RNAi 药物，将用于治疗家族性淀粉样多发性神经病变（hereditary transthyretin-mediated amyloidosis, hATTR; familial amyloidotic polyneuropathy (FAP)）。

2019年，药物GIVLAARI（givosiran）获得美国FDA 核准，成为第二支获准的RNAi药物，用于治疗成人急性肝性紫质症（acute hepatic porphyria, AHP）^[41]。

2020年，药物OXLUMO（lumasiran）获得美国FDA核准，用于治疗原发性高草酸尿症第1型( Primary Hyperoxaluria Type 1, PH1 )。^[42]
